【摘要】:Thismethodallowsfortherecoveryandenumerationofviableandculturable,non-tuberculosis,rapidlygrowingMycobacteria(M.immunogenum,M.chelonae,M.absessus,M.fortuitum,andM.smegmatis)inthepresenceofhighgramnegativebackgroundpopulationsinmetalworkingfluidfieldsamples.Duringthepastdecadeithasbecomeincreasinglyapparentthatnon-tuberculousMycobacteriaarecommonmembersoftheindigenousMWFbacterialpopulation.Thispopulationispredominantlycomprisedofgramnegativebacteriaandfungi.Mycobacterialcontaminationofmetalworkingfluidshasbeenputativelyassociatedwithhypersensitivitypneumonitis(HP)amongstmetalgrindingmachinists.Thedetectionandenumerationoftheseorganismswillaidinbetterunderstandingofoccupationalhealthrelatedproblemsandabetterassessmentofantimicrobialpesticideefficacy.Themeasurementofviableandculturablemycobacterialdensitiescombinedwiththetotalmycobacterialcounts(includingviableculturable(VC),viable-nonculturable(VNC)andnonviable(NV)counts)isusuallythefirststepinestablishinganypossiblerelationshipbetweenMycobacteriaandoccupationalhealthconcerns(forexample,HP).Themethodcanbeemployedinsurveystudiestocharacterizetheviable-culturablemycobacterialpopulationdensitiesofmetalworkingfluidfieldsamples.Thismethodisalsoapplicableforestablishingthemycobacterialresistanceofmetalworkingfluidformulationsbydeterminingmycobacteriumsurvivalbymeansofplatecounttechnique.ThismethodcanalsobeusedtoevaluatetherelativeefficacyofmicrobicidesagainstMycobacteriainmetalworkingfluids.1.1ThistestmethodcoversthedetectionandenumerationofviableandculturablerapidlygrowingMycobacteria(RGM),ornon-tuberculosisMycobacteria(NTM)inaqueousmetalworkingfluids(MWF)inthepresenceofhighnon-mycobacterialbackgroundpopulationusingstandardmicrobiologicalculturemethods.1.2Thedetectionlimitisonecolonyformingunit(CFU)/mLmetalworkingfluid.1.3ThistestmethodinvolvescultureoforganismsclassifiedasLevel2pathogens,andshouldbeundertakenbyatrainedmicrobiologistinanappropriatelyequippedfacility.ThemicrobiologistshouldalsobecapableofdistinguishingthediversecoloniesofMycobacteriafromothermicroorganismcoloniesonaPetridishandcapableofconfirmingMycobacteriabyacidfaststainingmethodThisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
